چکیده

Hydrogen peroxide (H2O2) is an important signaling molecule that involved in multiple physiological metabolic processes plants. Excess H2O2 can destroy biological macromolecules to poison the cell. Thioredoxin peroxidase (Tpx) plays role protecting plants from oxidative damage by clearing H2O2. In this study, tomato Tpx (SlTpx) gene was cloned and bioinformatic analysis done. The mRNA transcript level of SlTpx root leaf increased significantly after NaCl stress treatment for 12 h. overexpression transgenic tobacco were obtained study its function under stress. seed germination rate higher than wild type (WT) treatment. malondialdehyde (MDA) content reactive oxygen species (ROS) accumulation less WT Transgenic had antioxidant enzyme activities, proline total soluble sugar contents, expression Na+ metabolism genes WT. Moreover, seeds showed tolerance methyl viologen (MV) treatment, compared with Besides, growth prokaryotic strain pET-28a-SlTpx better pET-28a above results indicate improves plant salt scavenging Overexpression enhances tolerance.

Assessment of salt tolerance in transgenic tobacco (Nicotiana tobacum L.) plants expressing the AUX gene

Transformation of plants using Agrabacterium rhizogenes may affect secondary metabolite production as well as morphological changes. In this study, T-DNA from Ri plasmid in A. rhizogenes carrying pRi15834-PRT35S-GUS was introduced into tobacco leaf segments to initiate development of transformed hairy roots. Plant regeneration from transgenic roots used MS medium, and plants regenerated fro...

The yeast HAL1 gene improves salt tolerance of transgenic tomato.

Overexpression of the HAL1 gene in yeast has a positive effect on salt tolerance by maintaining a high internal K(+) concentration and decreasing intracellular Na(+) during salt stress. In the present work, the yeast gene HAL1 was introduced into tomato (Lycopersicon esculentum Mill.) by Agrobacterium tumefaciens-mediated transformation. A sample of primary transformants was self-pollinated, an...

Overexpression of a wheat aquaporin gene, TaAQP8, enhances salt stress tolerance in transgenic tobacco.

Aquaporin (AQP) proteins have been shown to transport water and other small molecules through biological membranes, which is crucial for plants to combat salt stress. However, the precise role of AQP genes in salt stress response is not completely understood in plants. In this study, a PIP1 subgroup AQP gene, designated TaAQP8, was cloned and characterized from wheat. Transient expression of Ta...

An Arabidopsis Mitochondrial Uncoupling Protein Confers Tolerance to Drought and Salt Stress in Transgenic Tobacco Plants

BACKGROUND Plants are challenged by a large number of environmental stresses that reduce productivity and even cause death. Both chloroplasts and mitochondria produce reactive oxygen species under normal conditions; however, stress causes an imbalance in these species that leads to deviations from normal cellular conditions and a variety of toxic effects. Mitochondria have uncoupling proteins (...

Overexpression of a Cytosolic Abiotic Stress Responsive Universal Stress Protein (SbUSP) Mitigates Salt and Osmotic Stress in Transgenic Tobacco Plants

The universal stress protein (USP) is a ubiquitous protein and plays an indispensable role in plant abiotic stress tolerance. The genome of Salicornia brachiata contains two homologs of intron less SbUSP gene which encodes for salt and osmotic responsive USP. In vivo localization reveals that SbUSP is a membrane bound cytosolic protein. The role of the gene was functionally validated by develop...
